Cytologists at the beginning of the twentieth century believed that the only way to understand how the living cell protoplasm worked was to study the chemical reactions that underpinned its functions. The dilemma was that to do so required cell structure to be destroyed, making it, they believed, impossible to have confidence that the reactions discovered in such dismembered and dead cells represented those of the intact, living cell. The resolution of this dilemma came from an integrated research strategy that used a morphologically guided and systematic disassembly of the cell into its parts and the biochemical analysis of those parts.
