Colleagues
we are attempting to create a 3D atlas of Xenopus embryos using the
light sheet scan of fixed and cleared embryos. The primary purpose is to
get an accurate cell count at various stages, but it would be nice to get
extra information of course, so I would love to know whether you have some
clues for us regarding various ways to stain the embryos to distinguish the
cell cycle phases, but also to estimate the time since the last cell
division. Any other clues regarding fixing, clearing or light sheet
scanning of the Xenopus embryos are most welcome. I'd also appreciate any
estimates of the number of cells you might already have. We so far
processed a single stage (11.5 ?) and counted ~40K cells total.
